"Do" is used with plural subjects and the pronouns I, you, we, and they (I do, you do, we do, they do). "Does" is used with singular third-person subjects: he, she, it, and singular nouns (he does, she does, the cat does, John does). In questions and negatives, the same rule applies: "Do you understand?" vs. "Does she understand?" "They don't know" vs. "He doesn't know."
